چکیده

1. Colchicine and vincristine only slightly inhibit the metabolism of glucose to CO(2) and lipids by isolated fat-cells. 2. Prolonged incubation with these agents causes no further inhibition. 3. Cytochalasin B, however, inhibits glucose metabolism to both CO(2) and lipids in fat-cells. 4. However, at a concentration that causes a strong inhibition of glucose metabolism cytochalasin B is without effect on the metabolism of pyruvate, lactate or arginine to these end products. The uptake of labelled alpha-aminoisobutyrate is likewise not modified. Similarly it does not affect release of glycerol or free fatty acid, or the actions of adrenaline, insulin or caffeine on these parameters. At 10mug/ml it slightly lowers ATP concentrations, an effect that does not occur at 2mug/ml. 5. The transport of fructose into adipocytes by a specific fructose-transport system is also not affected by the agent, but the uptake of 2-deoxyglucose is strongly inhibited. It is concluded that cytochalasin B may specifically inhibit the glucose-transport system of isolated fat-cells. 6. Cytochalasin A has a much weaker action than cytochalasin B on glucose metabolism.

Micropipette aspiration of human platelets: influence of microtubules and actin filaments on deformability.

The deformability of human platelets has been evaluated by micropipette aspiration. Control discoid platelets were about ten times as resistant to deformation in the micropipette as red blood cells. Under a constant negative pressure of 10 cm H2O, control platelets developed extension lengths of 0.74 +/- 0.1 micron. Prior treatment with vincristine, colchicine, or low temperature, all of which ...

Effect of Cytochalasin B and Colchicine on Α-amylase Release from Rat Parotid Tissue Slices

We have recently reported (3) that cytochalasin B, which disrupts microfilaments, and colchicine, which disrupts microtubules, interfered with the release of a-amylase from rat parotid tissue slices . Our observations on the effect of cytochalasin B on a-amylase release have been confirmed by Rifkin et al . (14) . Inhibitory actions of cytochalasin B or colchicine in several other systems have ...

Modulation of the effect of colchicine and cytochalasin B on fibroblast aggregates by the substratum.

In previous investigations, we have shown that aggregates of normal human fibroblasts displayed a remarkable number of morphogenetic properties in suspension and on a substratum. The present investigation has attempted to illustrate more clearly the role of colchicine- and cytochalasin-sensitive structures in the observed ultrastructural properties of the fibroblasts in this system. Cytochalasi...

Contractile proteins participate in release of erythroid growth regulators from mononuclear cells.

We have investigated the role of contractile proteins of circulating mononuclear cells in generation of membrane-associated, erythroid growth regulatory molecules. Lymphocytes and monocytes were incubated under serum-free conditions without and with cytochalasin B, cytochalasin D, or colchicine, and effects on positive and negative erythropoietic activities were determined in cell membranes and...

Human lymphocyte motility: normal characteristics and anomalous behavior of chronic lymphocytic leukemia cells.

The characteristics of human lymphocyte motility and its relationship to the redistribution of surface membrane antigens (capping) are poorly defined. Since chronic lymphocytic leukemia (CLL) cells cap poorly when compared with normal human lymphocytes, this study was undertaken to compare the motility of these two cell types. A modification of the Boyden chamber system was employed to quantify...
